Click the icon on the top right to purchase a tower. Click again to place the tower. Can only place the tower on the grass. (Green) Cannot place the tower on the dirt where the enemies are attacking. (Brown) After you have placed your tower on the grass (Green) you can click on the tower again to upgrade your tower. To upgrade your tower you need to earn money by defeating the enemies with your towers. The higher the cost the better the tower.
