Watching the in-game tutorials will help to understand where to position your hand. In this game you play the piano using the keyboard. Your hand should normally be positioned with your thumb over the X key, and your pointer finger over the D key. This lets you play "middle C" using your thumb like on a piano, and to me felt the most natural on a computer keyboard. The game also has a space mode that plays exactly the same way but with UFOs instead of notes, and lasers and explosions, just for fun.
