To begin the waves, press Begin, to Upgrade the tower, Press Upgrade and say "yes". To aim, use the mouse pointer. if your Money is higher than the number on your tower. you can press "Upgrade" and type in "Yes" in order to upgrade your tower. Note that price increases every upgrade.
